Enhanced Detection and Confirmation in Forensic Toxicology Screening Using a UHPLC-QTOF System

Accurate and reliable forensic toxicology screening of human body fluids for the presence of drugs, toxicants, poisons, and their metabolites is well known to be a challenging task. It is paramount that forensic data be of sufficient quality to stand up to rigorous scrutiny within a court of law. The Recreational Drugs European Network has discovered that over 650 new “designer” drugs have appeared in Europe over the past decade, and that number is continuing to rise. Because of its sensitivity, selectivity and wide scope of compound coverage, liquid chromatography–mass spectrometry (LC-MS) is now considered to be the technique of choice for forensic toxicological screening. Traditional LC-MS methods can suffer from high false positive rates because of complex matrix interferences or have challenges differentiating isobaric or isomeric compounds. In this presentation, we describe the use of enhanced detection and confirmation criteria with a UHPLC-QTOF workflow that exploits the use of “diagnostic ions” to reduce false positive findings, thus increasing the degree of confidence in correct compound identification. Key aspects discussed will include:

• High sensitivity forensic toxicology screening
• High confidence in compound identification
• Reduction of false positive detection rates
• Novel, comprehensive accurate mass forensic screening workflow
