Editors’ Series: HILIC: A Critical Evaluation

The basic principle of hydrophilic interaction liquid chromatography (HILIC) is to work with a polar stationary phase (bare silica or polar bonding) and a highly organic mobile phase (>70% acetonitrile). It is thus a powerful approach to retain polar compounds and it is also a good alternative to reversed-phase LC for the analysis of ionizable compounds of diverse polarity, often encountered in the pharmaceutical field. In addition, HILIC presents various advantages such as a higher ESI-MS sensitivity and a lower backpressure than in reversed-phase LC because of the higher volatility and weaker viscosity of the highly organic mobile phase.

In this web seminar, the possibilities and limitations of HILIC using columns packed with sub-2-µm fully porous particles and sub-3-µm core-shell particles will be discussed. The emphasis will be on kinetic performance, generic method development, the applicability of HILIC to a wide range of compounds (including polar compounds, ionizable compounds, and peptides) and a comparison of performance with reversed-phase LC. In addition, the critical impact of dissolution solvent in HILIC will be discussed and some problems related to pH control in highly organic mobile phase or long equilibrating time will be discussed.

Finally, HILIC-ESI-MS/MS will be compared with reversed-phase LC-ESI-MS/MS using a large number of compounds with very diverse physico-chemical properties, to mimic a multi-residue screening approach. Sensitivity and the possible ionization suppression or retention capability of HILIC and reversed-phase LC will be compared using ESI-MS/MS detection, to highlight the real benefits and limitations of a HILIC-ESI-MS/MS strategy.
