Raman Spectroscopy for Pharmaceutical Product Development and Manufacturing

In recent years, the availability of and improvements in process Raman instrumentation and the US Food and Drug Administration’s process analytical technology initiative have promoted the use of Raman spectroscopy as a technique to monitor pharmaceutical processes. Raman’s insensitivity to water and non-invasive nature makes Raman spectroscopy attractive as a routine technique. This session will discuss applications of Raman spectroscopy in pharmaceutical development as well as in the manufacturing of both active pharmaceutical ingredients and drug products. The capability of Raman as a spectroscopic tool as well as its ability to extract valuable information related to processes will be demonstrated via examples representing the monitoring of chemical reactions, polymorph conversion, solubilization and dissolution in liquids, determination of blend uniformity and potency, and polymorph conversion in solids. The usefulness of Raman data to determine the end point of processes and to establish potential correlations with process parameters and quality attributes will be presented. The challenges of method development, method transfer, and instrument/equipment interfacing will be highlighted.
